Recently, stem cells have been drawing increasing interest in basic and translational research that aims to understand stem cell biology and generate new therapies for various disorders. Many stem cells can be cultured in 2D relatively easily using tissue culture plastic. However, many of these cultures do not represent the natural conditions of stem cells in the body. In the body, microenvironments include numerous supporting cells and molecules. Therefore, researchers and clinicians have sought ideal stem cell preparations for basic research and clinical applications, which may be attainable through 3D culture of stem cells. The 3D cultures mimic the conditions of the natural environment of stem cells better, as cells in 3D cultures exhibit many unique and desirable characteristics that could be beneficial for therapeutic interventions. 3D stem cell cultures may employ supporting structures, such as various matrices or scaffolds, in addition to stem cells, to support complex structures. This book brings together recent research on 3D cultures of various stem cells to increase the basic understanding of stem cell culture techniques and also to highlight stem cell preparations for possible novel therapeutic applications.

Developed for a range of tissues where the culture environment takes into account the spatial organization of the cells therein, 3D cell culture models serve to bridge the gap between in vivo studies at one extreme with that of simple cell monolayers at the other. In 3D Cell Culture: Methods and Protocols, international experts describe a number of basic and applied methodologies taken from a breadth of scientific and engineering disciplines, many of which deal with direct applications of 3D culture models, most notably in the formation of tissues for clinical purpose. Beginning with an overview of the biological and materials scaffold requirements for successfully creating 3D models, the book delves into topics such as general scaffold design and fabrication techniques, models for bone, skin, cartilage, nerve, bladder, and hair follicles, and chapters on bioreactor design, imaging, and stem cells. Written in the highly successful Methods in Molecular BiologyTM series format, chapters include brief introductions to their respective subjects, lists of the necessary materials, step-by-step, readily reproducible laboratory protocols, and notes on troubleshooting and avoiding known pitfalls. Authoritative and cutting-edge, 3D Cell Culture: Methods and Protocols serves as a basic manual for laboratory-based scientists who not only need to have a comprehensive range of techniques contained within a single text but also require techniques described using a standard, convenient format.

A 3D stem cell culture is an artificially created environment that allows biological cells to grow or interact with surrounding extracellular environment in three dimensions. It can be classified into two types of methods, namely, scaffold techniques and scaffold-free techniques. Solid scaffolds, hydrogels, and other materials are used in scaffold techniques. Recent studies have explored the potentiality of human CD34+ stem cells generated in-vitro agarose gel 3D model to understand the process of bone ossification. Scaffold-free techniques include the use of low adhesion plates, hanging drop plates, micropatterned surfaces, rotating bioreactors, magnetic levitation, and magnetic 3D bioprinting. This book presents recent researches on 3D stem cell culture. Scientists and students actively engaged in the field of stem cell biology will find it full of crucial and unexplored concepts.
