Membrane fusion is an important molecular event which plays a pivotal role in many dynamic cellular processes, such as exocytosis, endocytosis, membrane biogenesis, fertilization, etc. While many reports on the physico-chem1cal process involved in membrane fusion have appeared in the literature and much information has accumulated, there has been no comprehensiv~ meeting of workers in the field. A recent symposium which took place at the Center for Tomorrow, State University of New York at Buffalo, New York, April 27-29, 1987, was the first meeting of its kind to specifically address the molecular mechanisms of membrane fusion. The Symposium consisted of oral, workshop and poster presentation sessions. The papers presented in the oral and workshop sessions are collected here and arranged approximately in the order presented at the Symposium. These papers are the most up-to-date and representative work at the for front of each aspect of membrane fusion. Although the readers may find some differences in interpretations regarding the molecular mechanisms of membrane fusion, there is an over all concensus that increased hydrophobicity and dehydration of the membrane surface are essential physico-chemical factors for membrane fusion to occur. Membrane fusion and targeting processes are tightly regulated and coordinated. Dozens of proteins, originating from both the cytoplasm and membranes are involved. The discovery of homologous proteins from yeast to neurons validates a unified view. Although much is known about the interfering proteins, the events occurring when two lipid bilayers actually fuse are less clear. It should be remembered that lipid bilayers behave like soap-bubbles fusing when meeting each other. In this respect interfering proteins should be considered as preventing undesirable and unnecessary fusion and eventually directing the biological membrane fusion process (when, where, how, and overcoming the activation energy). Over the last decade the volume Membrane Fusion. edited by Poste and Nicholson, has probably served as one of the major sources of review in formation on fusion in membrane systems. Since its publication much new information has been collected. New methods of inducing fusion have been invented or discovered, and new applications for fusion have been found. The need for an up-to-date monograph that covers and in tegrates these subjects, reviews established material, and rationalizes and integrates the old and the new is thus obvious. This book is the product of efforts to meet this need. Most of the current work in the field of membrane fusion takes place within the context of intact or modified cells. Hence this book emphasizes the plasma membrane.
