Translational Neuroimaging: Tools for CNS Drug Discovery, Development and Treatment combines the experience of academic, clinical and industrial neuroimagers in a unique collaborative approach to provide an integrated perspective of the use of small animal and human brain imaging in developing and validating translational models and biomarkers for the study and treatment of neuropsychiatric disorders. Translational Neuroimaging: Tools for CNS Drug Discovery, Development and Treatment examines the translational role of neuroimaging in model development from preclinical animal models, to human experimental medicine, and finally to clinical studies. The focus of this book is to identify and provide common endpoints between species that can serve to inform both the clinic and the bench with the information needed to accelerate clinically-effective CNS drug discovery. Traumatic brain injury (TBI) remains a significant source of death and permanent disability, contributing to nearly one-third of all injury related deaths in the United States and exacting a profound personal and economic toll. Despite the increased resources that have recently been brought to bear to improve our understanding of TBI, the development of new diagnostic and therapeutic approaches has been disappointingly slow. Translational Research in Traumatic Brain Injury attempts to integrate expertise from across specialties to address knowledge gaps in the field of TBI. Its chapters cover a wide scope of TBI research in five broad areas: Epidemiology Pathophysiology Diagnosis Current treatment strategies and sequelae Future therapies Specific topics discussed include the societal impact of TBI in both the civilian and military populations, neurobiology and molecular mechanisms of axonal and neuronal injury, biomarkers of traumatic brain injury and their relationship to pathology, neuroplasticity after TBI, neuroprotective and neurorestorative therapy, advanced neuroimaging of mild TBI, neurocognitive and psychiatric symptoms following mild TBI, sports-related TBI, epilepsy and PTSD following TBI, and more. The book integrates the perspectives of experts across disciplines to assist in the translation of new ideas to clinical practice and ultimately to improve the care of the brain injured patient.

Despite substantial progress in the development of neuroimaging methodologies, translational applications of neuroimaging remain scarce. This Research Topic invites article submissions that present promising neuroimaging applications and methods addressing critical needs for improving health outcomes. These may include Original Research, Clinical Trial, Systematic Review or Methods articles that investigate neuroimaging metrics as outcome measures or in combination with neural perturbation techniques (e.g., neurofeedback, neurostimulation), other translational applications (e.g., guiding neurosurgery). To foster debate, we also welcome critical Review, Opinion, and Perspective articles that survey the field and its progress towards clinical utility.

This book covers wide areas of animal and human psychopharmacology with clinical utility in the treatment of psychiatric and neurological (e.g Alzheimer's disease) disorders. The main theme is to develop a new paradigm for drug discovery that questions the claim that animal models or assays fail adequately to predict Phase 3 clinical trials. A new paradigm is advocated that stresses the importance of intermediate staging points between these extremes that depend on suitable translation of findings from animal studies to Phase 1 or Phase 2 studies utilising experimental medicine.

The sophisticated methods of neuroscience—including molecular genetics, structural and functional neuroimaging, animal models, and experimental tasks that approximate real-world behaviors in human research—have yielded important insights about typical functioning and neurobehavioral disorders. Translational neuroscience endeavors to use this knowledge to improve the human condition by developing and improving interventions for these disorders. This paper reviews the literature on the contribution of functional magnetic resonance imaging (fMRI) and two related techniques, resting-state fMRI (rs-fMRI) and real-time fMRI (rt-fMRI), to the diagnosis and treatment of behavioral problems and psychiatric disorders. It also explains how incorporating neuroscience principles and techniques into research on the prevention of substance misuse and antisocial behavior may spur advances and innovations in this important area. This article argues that fMRI’s potential contribution to these prevention efforts has yet to be fully realized, explores new ways in which the technique could be adapted to that end, highlights some of the work by researchers in the vanguard of this effort, and notes limitations of fMRI and ethical concerns the technique raises.
