Many methods have been developed to provide thermodynamic measurements of protein stability. These measurements are of great importance as they are widely used in biophysical studies of protein folding and function. Recently, an amide hydrogen exchange- and matrix-assisted laser desorption/ionization (MALDI) mass spectrometry-based method, termed SUPREX (stability of unpurified proteins from rates of H/D exchange), was reported to make such thermodynamic measurements. The SUPREX technique removes some important experimental limitations associated with conventional spectroscopic and calorimetric methods for making similar thermodynamic measurements. This dissertation is focused on the application of SUPREX technique to several new protein systems.
