The Handbook of Computational Neurodegeneration provides a comprehensive overview of the field and thus bridges the gap between standard textbooks of research on neurodegeneration and dispersed publications for specialists that have a narrowed focus on computational methods to study this complicated process. The handbook reviews the central issues and methodological approaches related to the field for which the reader pursues a thorough overview. It also conveys more advanced knowledge, thus serving both as an introductory text and as a starting point for an in-depth study of a specific area, as well as a quick reference source for the expert by reflecting the state of the art and future prospects. The book includes topics that are usually missing in standard textbooks and that are only marginally represented in the specific literature. The broad scope of this handbook is reflected by five major parts that facilitate an integration of computational concepts, methods and applications in the study of neurodegeneration. Each part is intended to stand on its own, giving an overview of the topic and the most important problems and approaches, which are supported by examples, practical applications, and proposed methodologies. The basic concepts and knowledge, standard procedures and methods are presented, as well as recent advances and new perspectives.

Neurodegenerative Disorders provides an accessible guide to the commonest neurodegenerative diseases, outlining the main clinical features, treatment options and outcomes of the conditions most frequently encountered in clinical practice. Beginning with an overview of the general principles that underlie degeneration, and the contribution of established and new diagnostics techniques, the book goes on to describe the most common neurodegenerative conditions including Alzheimer Disease; Dementia and Cerebrovascular Disease; Parkinson’s Disease; Fronto-Temporal Dementia; Amyotrophic Lateral Sclerosis; Huntington’s Disease; Parkinsonism Plus syndromes and Prion Diseases. Final chapters cover important management issues including the use of palliative care strategies, advance directives and competence. Written by experts in the field internationally Neurodegenerative Disorders is a practical guide for clinicians that will be indispensable for the management of these conditions.

Collectively, neurodegenerative diseases are characterized by chronic and progressive loss of neurons in discrete areas of the brain, producing debilitating symptoms such as dementia, loss of memory, loss of sensory or motor capability, decreased overall quality of life eventually leading to premature death. Two types of cell death are known to occur during neurodegeneration: (a) apoptosis and (b) necrosis. The necrosis is characterized by the passive cell swelling, intense mitochondrial damage with rapid loss of ATP, alterations in neural membrane permeability, high calcium influx, and disruption of ion homeostasis. This type of cell death leads to membrane lysis and release of intracellular components that induce inflammatory reactions. Necrotic cell death normally occurs at the core of injury site. In contrast, apoptosis is an active process in which caspases (a group of endoproteases with specificity for aspartate residues in protein) are stimulated. Apoptotic cell death is accompanied by cell shrinkage, dynamic membrane blebbing, chromatin condensation, DNA laddering, loss of phospholipids asymmetry, low ATP levels, and mild calcium overload. This type of cell death normally occurs in penumbral region at the ischemic injury site and in different regions in various neurodegenerative diseases.
