Mass spectrometry has emerged as a powerful tool for analyzing and identifying biomolecules of interest, including small molecules, pharmaceutical compounds, and tissue biomarkers - such as proteins and glycans. While most mass spectrometry based analytical techniques focus on liquid-chromatography coupled with tandem mass spectrometry for biomolecule analysis, mass spectrometry imaging (MSI) provides a unique advantage to map spatial distribution within an anatomical context of the tissue of interest. We use a high-resolution accurate mass MALDI Orbitrap LTQ XL mass spectrometer, that offers enhanced mass spectral resolution and high accuracy mass measurements critical for molecular identification, to perform MSI on thin tissue sections. In this thesis, I will highlight specific examples where the information gained from imaging is critical to understanding the question of interest: 1) Development of pathology-directed biomarker analysis of clinically relevant tissues 2) Pushing throughput for MSI in vitro drug penetration analysis of three-dimensional cell cultures including spheroid and organoid systems and 3) Analysis of lipids in insulator regions of the electric eel for the development of bioelectric batteries.
