Detailed protocols for analyzing DNA strand breaks, specific RNA/DNA expressions, and DNA binding proteins such as transcription factors at the individual cell level. It describes in situ nick translation and TUNEL in detail, along with radioactive and non-radioactive methods for in situ hybridization. Of special significance is the description of Southwestern histochemistry, which makes it possible to analyze the expression of transcription regulatory factors in individual cells. This volume provides an overview of the current status of molecular histochemistry along with practical how-to-do-it details. The methodology is set out in easy-to-follow language by leading scientists working in the field today. Both a rich source of the latest information and a practical lab manual.

This detailed volume explores numerous histochemical techniques through a series of lab-tested protocols for the detection of specific molecules or metabolic processes, both at light and electron microscopy. More in detail, the book is divided into six sections covering a variety of chemical targets. It begins with a section on vital histochemistry and continues with chapters on histochemistry as it relates to lectins, proteins, lipids, DNA and RNA, as well as plants. The volume also contains four overview chapters on vital histochemistry, lectin histochemistry, and DNA fluorochromes. Written for the highly successful Methods in Molecular Biology series, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and tips on troubleshooting and avoiding known pitfalls. Authoritative and practical, Histochemistry of Single Molecules: Methods and Protocols aims to effectively help scientists in very different research fields to elucidate biological issues though a unique approach to molecular biology in situ.

Toxicological and pharmacological effects arise when chemicals interact with biophysiological functions in discrete cell types. There is a continuing need to screen novel compounds for their potential therapeutic effects, and once these have been "discovered" to understand their molecular actions, as the basis of using such compounds safely and for rational drug design. Pharmacology now uses all of the sophisticated molecular research techniques that are available for the development of safer and more efficacious drugs. Histochemistry has been usefully applied to developing new drugs (and assessing chemical safley) and is potentially cost effective. The need to test novel substances for their potential adverse effects has raised many questions. Toxicological pathology has moved away from the cataloging of lesions towards understanding the basis of the events that underly cell injury, especially for those secondary consequences of chemical injury that lead to malignancy and chronic disease. The focal nature of toxicologic lesions de mands the use of microtechniques to provide data to help understand these questions. Histochemistry is under-utilized, but offers one of the key ap proaches necessary to address the problem of understanding interactions between a cell population and a chemical, the modulation of cellular biochem istryor the presence of a lesion in a test animal can be rationalised in terms of species differences that have no relevance to man as opposed to those that are of clinical significance or represent a warning of dire consequences to man.

This fifth edition of Histological and Histochemical Methods continues to provide a clear and consistent introduction to the techniques, description and analysis of the chemical and physical principles of fixation, tissue processing, staining, enzyme location, immunohistochemistry and other key procedures. The overall structure of the book remains unchanged, but the content has been heavily revised to update the techniques used in line with recent technological advances. Additionally, there are new sections on: Artefacts and troubleshooting Methods for microorganisms and fungi in sections Methods for various pigments and mineral deposits in tissues Methods for skeletal elements (bone, cartilage) in whole-mounts Histological and Histochemical Methods 5e is essential reading for students, lecturers, researchers and professionals using histological and histochemical techniques. From reviews: "Histological and Histochemical Methods is a tour de force wholly suited to the modern age of histology and Professor Kiernan has triumphed again. To cover so much ground clearly and concisely while including the justification of the underlying chemistry makes this book unique. There should not be a histology laboratory or an undergraduate library that does not own a copy." Biotechnic & Histochemistry 2016, 91(2): 145. "This book should be present on the bookshelves of every research or analysis laboratory where histology and histochemistry are routinely used, as an essential reference source of basic and practical information for scientists and technicians." European Journal of Histochemistry, 2016, vol. 60.

Somuchofwhatweknowaboutthepathogenesisofhumandiseasehascomefromthe systematic and careful study of histological material. Indeed, every internal medicine discipline has its landmark papers describing the clinico-pathological correlations. However, increasingly, it is molecular and cellular biology that provides the necessary mechanistic insights. For many years, it was thought that the two skill sets were mutually exclusive, but we hope that this book shows that this is not necessarily so. Implicitinthescienceofhistologyisthepreservationandarchivingoftissue.PartIof the book concentrates on the preparation of tissue, providing an overview of fixation, embedding, and processing (Chapter 1), and in Chapters 2 and 3, the required techniques for the retrieval of RNA from histological sections. Both routine and specialist histological staining techniques are provided in Part II. These include pro- cols for immuno (Chapters 4–7), lectin (Chapter 8), and hybridization (Chapter 9) histochemistry, histologicalstaining (Chapters10and11),aswellasspecificmethods for the in situ identification of hypoxia (Chapter 12) and apoptosis (Chapter 13). Finally, Part III details advances in imaging (Chapters 14–16) and image analysis (Chapter 17). It is hoped that this volume will provide molecular biologists with the basic his- chemical techniques and histologists with the molecular techniques to realise the potential of their resource. We are indebted to the authors for their generosity in sharing these protocols.
