The interaction of neurotransmitters, neuromodulators and neuroactive drugs with receptors localized at the cell surface initiates a chain of molecular events leading to integrated neuronal responses to the triggering stimuli. Major advancements in the characterization and isolation of recep tor molecules have answered many quest ions regarding the nature of the ele ments that determine the specificity in these interactions. At the same time, recent studies have provided evidence that delicate regulation by intracellular enzymatic systems determines the efficiency of the stimulus response coupling process, mediates the interaction between receptors, operates in feedback control mechanisms and transduces signals from the receptors to various effector sites in a highly coordinated fashion. These studies are at the focus of the present volume, which is an outcome of a symposium held at the University of Vermont College of Medicine on March 21-23, 1986, in conjunction with the seventeenth annual meeting of the Amer ican Society for Neurochemistry. The symposium has demonstrated clearly that the concerted efforts of investigators in neurophysiology, biochemis try, pharmacology, cell-biology, molecular genetics, neurology, and psy chiatry are required to achieve better understanding of the processes under lying neuronal responsiveness. This volume includes contributions provided by prominent investigators in all these research areas. We hope that the readers will find here a useful source of information and ideas for stimu lating further studies which may serve to narrow the gap between basic neuroscience research and its clinical implications.

The last decades have witnessed a radical change in our views on central nervous system damage and repair. This change is not only due to the emergence of new powerful tools for the analysis of the brain and its reactions to insults, but it also reflects a conceptual change in the way we approach these problems. As an illustration to this development, it is instructive to go back to the proceedings of a meeting at the NIH in 1955 edited by William F. Windle, which summarizes the disillusioned and pessimistic view on CNS regeneration prevailing at the time. While this generation of researchers were well aware of the issues at stake, they felt they had reached the end of the road; the approaches they had pursued had got stuck and the tools available could not take them any further. I can very well imagine that the participants, most of them leaders in the field, left that conference feeling they had heard their field being sentenced to death.

Numerous studies have proven the biological basis of memory formation and have begun to identify the biochemical traces and cellular circuits that are formed by experience, and which participate int the storage of information in the brain, its retention for long durations, and its retrieval upon demand. Cells in the nervous system have the capability of undergoing extremely long-lasting alterations in response to hormonal, pharmacological, and environmental stimulations. The mechanisms underlying this neuronal plasticity are activated by experiential inputs and operate in the process of learning and the formation of memories in the brain. This volume presents research areas which have not been highlighted in the past. In addition to studies on the involement of functional proteins in neuronal adaptation, this volume presents recent developments on the critical roles of bioactive lipids and nucleotides in these processes. In addition to the widely studied role of second messengers, a review of studies on extracellular phosphorylation systems operating on the surface of brain neurons is presented.The first section of the volume presents studies of basic mechanisms operating in a wide range of adaptive processes. The second section presents recent advances in investigations that have demonstrated the clinical implications of this research. These include: state of the art use of transgenic models in studies of molecular and cellular mechanisms implicated in familial Alzheimer's disease and Amyotrophic Lateral Sclerosis; studies of specific proteins implicated in Alzheimer's disease, including an adapter that binds to the beta-amyloid precurser protein (beta-APP) and the microtubular protein Tau and its membrane-bound counterpart.
