Skin Tissue Models provides a translational link for biomedical researchers on the interdisciplinary approaches to skin regeneration. As the skin is the largest organ in the body, engineered substitutes have critical medical application to patients with disease and injury – from burn wounds and surgical scars, to vitiligo, psoriasis and even plastic surgery. This volume offers readers preliminary description of the normal structure and function of mammalian skin, exposure to clinical problems and disease, coverage of potential therapeutic molecules and testing, skin substitutes, models as study platforms of skin biology and emerging technologies. The editors have created a table of contents which frames the relevance of skin tissue models for researchers as platforms to study skin biology and therapeutic approaches for different skin diseases, for clinicians as tissue substitutes, and for cosmetic and pharmaceutical industries as alternative test substrates that can replace animal models. Offers descriptions of the normal structure/function of mammalian skin, exposure to clinical problems, and more Presents coverage of skin diseases (cancer, genodermatoses, vitiligo and psoriasis) that extends to clinical requirements and skin diseases in vitro models Addresses legal requirements and ethical concerns in drugs and cosmetics in vitro testing Edited and authored by internationally renowned group of researchers, presenting the broadest coverage possible

3D tissue modelling is an emerging field used for the investigation of disease mechanisms and drug development. The two key drivers of this upsurge in research lie in its potential to offer a way to reduce animal testing with respect to biotoxicity analysis, preferably on physiology recapitulated human tissues and, additionally, it provides an alternative approach to regenerative medicine. Integrating physics, chemistry, materials science, and stem cell and biomedical engineering, this book provides a complete foundation to this exciting, and interdisciplinary field. Beginning with the basic principles of 3D tissue modelling, the reader will find expert reviews on key fabrication technologies and processes, including microfluidics, microfabrication technology such as 3D bioprinting, and programming approaches to emulating human tissue complexity. The next stage introduces the reader to a range of materials used for 3D tissue modelling, from synthetic to natural materials, as well as the emerging field of tissue derived decellularized extracellular matrix (dECM). A whole host of critical applications are covered, with several chapters dedicated to hard and soft tissues, as well as focused reviews on the respiratory and central nervous system. Finally, the development of in vitro tissue models to screen drugs and study progression and etiologies of diseases, with particular attention paid to cancer, can be found.

This volume of Current Problems in Dermatology presents the reader with a portrait of the scientific background of the complex process of safety assessment of cosmetics as well as information on European cosmetic legislation. A practical approach to the search for toxicity data on cosmetic ingredients and a compilation of the legally required technical dossier of a finished cosmetic product as well as an in-depth analysis of the safety assessment of cosmetic ingredients performed at the EU level by the Scientific Committee on Consumer Products (SCCP) are available. The current status of 3R alternatives to animal testing and the extent to which they are implemented by the cosmetic industry and considered by the SCCP in the risk assessment process is discussed. As such, guidance is provided in relation to the manifold challenges cosmetic safety assessors are faced with in the current EU regulatory setting. In this valuable handbook, qualified cosmetic safety assessors, suppliers of raw materials, dermatologists and pharmacists, toxicologists as well as EU officials and administrators dealing with cosmetics will find relevant information on the European cosmetic legislation, the compilation of cosmetic technical dossiers, toxicological database searches, and the availability and use of alternative methods in the field of cosmetics.

Since its advent, nanotechnologies are considered key enabling technologies that take advantage of a wide array of nanomaterials (NMs) for biomedical and industrial applications generating significant societal and economic benefits. However, such innovation increases human exposure to these substances through inhalation, ingestion or dermal contact raising public health concerns. Furthermore, the NMs’ specific physicochemical properties, that confer them unique beneficial characteristics, can also elicit nano-bio interactions leading to toxicity and concerns for public health. In addition, such properties can be affected by the surrounding matrix, particularly when incorporated in complex matrices such as food products, leading to secondary features potentially more relevant than primary characteristics for determining their toxicological outcome. These nano specific issues raise the question of whether the NMs may produce adverse outcomes that are not accounted for when using conventional toxicological approaches to assess their safety. Such uncertainties about the safety of NMs for human health and the environment may hamper a faster and more widespread exploration of their potentials. In response, the NMs definition has evolved, and nanotoxicology has developed towards new and more integrative approach methods to support regulatory and policy actions. This book provides a perspective on recent developments in the synthesis, application, and characterization of NMs and the related nanotechnologies, focusing on nanotoxicology for their accurate safety assessment early in the product development stage. The use of complex in vitro models, including multicellular systems and organoids, and “omics-based” approaches, such as transcriptomics or epigenomics, have greatly contributed to an in-depth understanding of the cellular and molecular mechanisms behind some NMs toxicity. Such mechanistic knowledge is equally addressed in this book and has set the basis for a predictive nanotoxicology approach building on adverse outcome pathways. In addition, considering the knowledge provided by the above-mentioned approaches, insights into risk assessment, standardization, and regulation of NMs are also included. Incorporating adequate nanosafety assessment early in the life-cycle of NMs will allow the implementation of the safe and sustainable-by-design paradigm enabling safety to keep pace with innovation. The rapid expansion of the nanotechnology field raises concerns, like any new technology, about the toxicity and environmental impact of nanomaterials. This book addresses the gaps relating to health and safety issues of this field and aims to bring together fragmented knowledge on nanosafety. Not only do chapters address conventional toxicity issues, but also more recent developments such as food borne nanoparticles, life cycle analysis of nanoparticles and nano ethics. In addition, the authors discuss the environmental impact of nanotechnologies as well as safety guidelines and ethical issues surrounding the use of nanoparticles. In particular this book presents a unique compilation of experimental and computational perspectives and illustrates the use of computational models as a support for experimental work. Nanotoxicology: Experimental and Computational Perspectives is aimed towards postgraduates, academics, and practicing industry professionals. This highly comprehensive review also serves as an excellent foundation for undergraduate students and researchers new to nanotechnology and nanotoxicology. It is of particular value to toxicologists working in nanotechnology, chemical risk assessment, food science, environmental, safety, chemical engineering, the biological sciences and pharmaceutical research.
