Chemiluminescence immunoassay is now established as one of the best alternatives to conventional radioimmunoassay for the quantitation of low concentrations of analytes in complex samples. During the last two decades the technology has evolved into analytical procedures whose performance far exceeds that of immunoassays based on the use of radioactive labels. Without the constraints of radioactivity, the scope of this type of analytical procedure has widened beyond the confines of the specialist clinical chemistry laboratory to other disciplines such as microbiology, veterinary medicine, agriculture, food and environmental testing. This is the first work to present the topic as a subject in its own right. In order to provide a complete picture of the subject, overviews are presented of the individual areas of chemiluminescence and immunoassay with particular emphasis on the requirements for interfacing chemiluminescent and immunochemical reactions. The possible ways of configuring chemiluminescence immunoassays are described. State-of-the-art chemiluminescence immunoassay systems are covered in detail together with those systems which are commercially available. The book is aimed at researchers and routine laboratory staff in the life sciences who wish to make use of this high-performance analytical technique and also at those interested in industrial applications of the technology in the food, agricultural and environmental sciences.

Describes the occurrence, chemistry, and measurement of chemiluminescence, a phenomenon that together with the use of very sensitive light detectors has led to a number of biomedical applications and new concepts about disease processes. Covers the principles of using the reactions from fireflies and luminous bacteria to measure enzymes and metabolites; calcium-sensitive proteins from luminous jellyfish to measure changes in calcium inside living cells; the measurement of oxygen radicals and the replacement of radioactive labels in immunoassay by chemiluminescent compounds; and future applications in research and clinical laboratories. `In the second edition of Principles I have attempted to maintain the emphasis on basics, while updating the examples to include more recent results from the literature. There is a new chapter providing an overview of extrinisic fluorophores. The discussion of timeresolved measurements has been expanded to two chapters. Quenching has also been expanded in two chapters. Energy transfer and anisotropy have each been expanded to three chapters. There is also a new chapter on fluorescence sensing. To enhance the usefulness of this book as a textbook, most chapters are followed by a set of problems. Sections which describe advanced topics are indicated as such, to allow these sections to be skipped in an introduction course. Glossaries are provided for commonly used acronyms and mathematical symbols. For those wanting additional informtion, the final appendix contains a list of recommended books which expand on various specialized topics.' from the author's Preface

Molecular diagnostic procedures have been described in a number of recent books and articles. However, these publications have not focused on virus detection, nor have they provided practical protocols for the newer molecular methods. Written by the inventors or principal developers of these technologies, Molecular Methods for Virus Detection provides both reviews of individual methods and instructions for detecting virus nucleic acid sequences in clinical specimens. Each procedure includes quality assurance protocols that are often ignored by other methodology books. Molecular Methods for Virus Detection provides clinically relevant procedures for many of the newer diagnostic methodologies. Provides state-of-the-art PCR methods for amplification, quantitation, in situ hybridization, and multiplex reactions Goes beyond PCR with protocols for 3SR, NASBA, LCR, SDA, and LAT Covers important virus detection methods such as in situ hybridization; Southern, dot, and slot blots; branched chain signal amplification; and chemiluminescence Includes quality control information crucial in research and clinical laboratories Most chapters are written by the inventors and principal developers of the methodologies Includes color plates, 77 figures, and 18 tables

Discover how metal-enhanced fluorescence is changing traditional concepts of fluorescence This book collects and analyzes all the current trends, opinions, and emerging hot topics in the field of metal-enhanced fluorescence (MEF). Readers learn how this emerging technology enhances the utility of current fluorescence-based approaches. For example, MEF can be used to better detect and track specific molecules that may be present in very low quantities in either clinical samples or biological systems. Author Chris Geddes, a noted pioneer in the field, not only explains the fundamentals of metal-enhanced fluorescence, but also the significance of all the most recent findings and models in the field. Metal-enhanced fluorescence refers to the use of metal colloids and nanoscale metallic particles in fluorescence systems. It offers researchers the opportunity to modify the basic properties of fluorophores in both near- and far-field fluorescence formats. Benefits of metal-enhanced fluorescence compared to traditional fluorescence include: Increased efficiency of fluorescence emission Increased detection sensitivity Protect against fluorophore photobleaching Applicability to almost any molecule, including both intrinsic and extrinsic chromophores Following a discussion of the principles and fundamentals, the author examines the process and applications of metal-enhanced fluorescence. Throughout the book, references lead to the primary literature, facilitating in-depth investigations into particular topics. Guiding readers from the basics to state-of-the-technology applications, this book is recommended for all chemists, physicists, and biomedical engineers working in the field of fluorescence.

Time-resolved fluorescence spectroscopy is widely used as a research tool in bioch- istry and biophysics. These uses of fluorescence have resulted in extensive knowledge of the structure and dynamics of biological macromolecules. This information has been gained by studies of phenomena that affect the excited state, such as the local environment, quenching processes, and energy transfer. Topics in Fluorescence Spectroscopy, Volume 4: Probe Design and Chemical Sensing reflects a new trend, which is the use of time-resolved fluorescence in analytical and clinical chemistry. These emerging applications of time-resolved fluorescence are the result of continued advances in laser detector and computer technology. For instance, pho- multiplier tubes (PMT) were previously bulky devices. Miniature PMTs are now available, and the performance of simpler detectors is continually improving. There is also considerable effort to develop fluorophores that can be excited with the red/ne- infrared (NIR) output of laser diodes. Using such probes, one can readily imagine small time-resolved fluorometers, even hand-held devices, being used fordoctor’s office or home health care.
