Glutathione Centennial: Molecular Perspectives and Clinical Implications is a collection of papers presented at the international symposium by the same titles, held in Osaka, Japan on December 1-4, 1988. This symposium concentrates on the developments is glutathione field, especially those related to molecular and clinical problems. This book is organized into five parts encompassing 30 chapters. Part I deals with the metabolic regulation, thermodynamic and kinetic properties, and enzyme regulation of glutathione. Part II examines the gene expression and molecular mechanism of glutathione-related enzymes, while Part III explores the association between carcinogenesis, xenobiotic compounds, and glutathione. Part IV highlights the clinical implications of glutathione, particularly its effect on vasospasm in subarachnoid hemorrhage and experimental hepatitis. Part V discusses the mechanism and regulation of biological transport of glutathione. This is the first serious attempt to synthesize all that became known of glutathione over the last three decades. The book contains an update of glutathione biosynthesis with special emphasis on its regulation in adaptive stress responses. Other chapters review glutathione transport systems and glutathione peroxidases and their differences in substrate specificities and localization. Further contributions center on the diversified roles of different glutathione-S-transferases and the roles of nitrosoglutathione and glutaredoxins - a subfamily of redoxins. The book closes with discussions of the analogous or homologous thiol metabolism in pathogens and the potential suitability of involved enzymes as drug targets. The goal of this text is to focus readers attention on three major areas; the origin and localization of GSH in the nervous system; the multiple effects of GSH on neural health activity; and the potential for alterations on GSH status to lead to neurological damage of the type observed in amyotrophic lateral sclerosis, Parkinson's disease and other neurological disorders. The text also touches upon the additional roles of the antoxidant GSH, including possible neurotransmitter action, redox modulation of ionotropic receptor function, and neuroprotection against exicitoxic actions of glutamate.

Structure and Function of Glutathione S-Transferases provides some of the latest information available on a variety of structural and functional components of glutathione S-transferases, a family of isozymes involved in many endogenous and exogenous functions in cells. Molecular studies presented in the book focus on the regulation of these enzymes and identify important response elements. X-ray crystallographic structures show detailed information on the structural aspects of these proteins. Metabolism of a number of carcinogens and drugs is covered in detail, and the role that these enzymes play in governing drug resistance at the preclinical and clinical levels is discussed. This volume is a summary of the proceedings of the 8th International Symposium on Vitamin B6 and Carbonyl Catalysis, held in Osaka, Japan, 15-19 October 1990. The development of molecular biology and x-ray crystallography techniques have contributed greatly to the knowledge and understanding of PLP-dependent enzymes. The structural, mechanistic and functional aspects of the PLP-dependent enzymes have been studied for over 40 years by many active researchers. Recent evidence indicates that the coenzyme is substituted by the pyruvoyl group which is a reactive carbonyl in some amino acid decarboxylases, and the presence of pyrroloquinoline quinone (PQQ), another carbonyl coenzyme, in other enzymes. The contents of this volume attest to the exciting progress in elucidating structure, mechanism of action, and the function of enzymes dependent on coenzymes containing the carbonyl group.

Since the appearance of photosynthesis on our planet, all living organisms have been facing a new abundant, extremely reactive element, oxygen. This element is used for the synthesis of highly energetic compounds, but can also generate molecules capable of damaging constituents of living structures, including proteins, nucleic acids and lipids. It is therefore no surprise that all biological organisms have evolved multiple and sophisticated ways to reduce the detrimental effects of oxygen. For cells and tissues of highly organized animals, particularly cells of the immune system, superoxidation products play an important role, via oxidative stress, in activation, inflammation and viral and bacterial infections. We must be grateful to Drs. C. Pasquier, C. Auclair, L. Packer and R. Olivier for having brought together many specialists in an international meeting held in Paris in March 1993, at the Ministry of Research. This book comprises an impressive amount of recent knowledge, a real mine for the reader in this fast developing field of research. Furthermore, we should not forget that this basic research may lead in the future to new therapeutic approaches to the most important pathologies of the latter part of this century, including AIDS.
