This special article collection of Frontiers in Pharmacology includes reviews and original articles on different aspects of IL-1 inhibition. Since the time IL-1 and its natural antagonist IL-1Ra have been discovered, specific IL-1 targeted therapies have been developed to cure an increasing number of diseases. The purpose of this Research Topic is to provide an overview of the different clinical uses of IL-1 blockade and new insights in basic research issues.

Inflammation has become one of the most exciting and rewarding areas of medical research. Recent years have seen a revolution in our understanding of how blood and tissue cells interact and of the intracellular mechanisms controlling their activation. This has revealed the underlying inflammatory pathology of many diseases and provided multiple new targets for anti-inflammatory and immunomodulatory therapy. This living project will serve as a reliable and comprehensive data pool for everybody working in inflammation research. Owing to its dynamic nature, it will grow with time and future editions, becoming an indispensible source of information for academia, clinical practitioners and industry.

Cytokine Storm Syndromes, including HLH and MAS, are frequently fatal disorders, particularly if not recognized early and treated during presentation. The genetics of Cytokine Storm Syndromes are being defined with many of the risk alleles giving rise to mutations in the perforin-mediated cytolytic pathway used by CD8 cytotoxic T cells and natural killer cells. These are being studied using murine models. It is only during the last decade that the functions of sinusoidal endothelial cells, Kupffer cells, hepatic stellate cells, pit cells and other intrahepatic lymphocytes have been better understood. The development of methods for isolation and co-culturing various types of liver cells has established that they communicate and cooperate via secretion of various intercellular mediators. This monograph summarizes multiple data that suggest the important role of cellular cross-talk for the functions of both normal and diseased liver. Investigations of the activation, proliferation, and, in some cases, differentia tion of mononuclear cells involved in the immune response are proceeding rapidly. These studies have resulted in the discovery of several factors that promote these cellular events, some of which have been characterized biochemically to various extents. Because of the considerable interest in understanding these cellular changes at the molecular level, we chose to produce the first thematic volume for Contemporary Topics in Molecular Immunology; the theme deals with certain regulatory factors that promote proliferation and differentiation. We have com piled contributions from numerous scientists well known for their work with several regulatory factors. In the following paragraphs, the reader will find an overview of the contents of this volume. Greene and Robb review data they have generated over the past 2-3 years with respect to characterization of hormone-specific Interleukin-2 (IL-2) recep tors on the surface of activated T cells. Their chapter traces the development of a quantitative assay for assessment of IL-2 receptors based on the preparation and use of radiolabeled IL-2 prepared biosynthetically with the aid of IL-2-pro ducer leukemic cells. The authors then describe an alternate approach, the prep aration of a monoclonal antibody previously determined to be directed against a T -cell-activation antigen. This so-called anti-Tac antibody was later found to recognize a determinant on or near the IL-2 receptor.

This book is devoted to innovative medicine, comprising the proceedings of the Uehara Memorial Foundation Symposium 2014. It remains extremely rare for the findings of basic research to be developed into clinical applications, and it takes a long time for the process to be achieved. The inflammasome was first described in 2002 as a molecular complex activating proinflammatory caspases and therefore regulating the maturation and biological activities of cytokines such as IL-1 and IL-18. This finding was substantiated by the identification of several mutations in the cias1 gene, encoding the human NLRP3 protein, responsible for several autoinflammatory disorders such as the Muckle Wells syndrome. Since, the interest for this complex has constantly increased and several inflammasome complexes with different specificities have been described. These inflammasomes sense a wide variety of pathogens and danger signals and are key players in the inflammatory response. With the contributions of leading international experts in the field, this book provides an extensive overview of the current knowledge of inflammasome biology and their role in health and disease.
